Not all pellet guns use C02 some are designed to use a spring or pump. Pellet guns that use C02 are rapid fire guns and do not require re-cocking or pumping between each shot. BUT they usually have less power that spring or pump pellet guns.

Yes. In fact it's not recommended to leave a charged C02 pellet in a BB gun. It will eventually rupture the seals. After shooting release the C02 gas. If you have a pistol that requires a pellet to be in the handle so the grip can stay attached, then leave a discharged C02 pellet in the handle to keep the grip in place.
When the C02 cylinder is placed into the gun and screwed in the end of the cylinder is pierced and some C02 charges the system of the pistol. When the trigger is pulled a burst of the C02 gas is released behind the pellet, firing it down the barrel. You can usually get about 40 to 50 shots from as regular 12 oz C02 cylinder.
