If you're asking in regards to Super Smash Bros. Brawl, the "R" button is actually the "Z" button on the back of the nunchuck. Hold it down while you're picking your character, for example, Samus. Once you've chosen her, and are on your way to playing "Classic Mode" release it after you select "Start".
You will play with Zero Suit Samus then.
