There are a few theories, is was that The Joker once was an namless engineer that got fired from his job of a chemical plant to start a career as stand up comedian. He failed and got in finincial problems. Desprate to take care of his wife he agreed with to mobsters that offered him a part in a armed robbery on his old plant. He disquesed himself as "The Red Hood" to complete the illusion. He was than cornered by Batman, when he tried to tell him he wasn't bad, he fell into a vat of acid. The world belived him death, but he wasn't. The acid died his hair green and made his skin permanent white.
Another is that The Joker once was a criminal named J. Napier, who fell in acid.

At the age of six Bruce Wayne's parents are brutally murdered in a street robbery by a couple of criminals. During the robbery one of the assailants uses the phrase:
"Have you ever danced with the devil in the pale moonlight? I ask that of all my prey. I just like the sound of it."
The murder of Bruce's parents affects the boy and he makes a promise to make the criminals pay. When he grows up he discovers caves in the Wayne stately home inhabited by Bats. This gives him the idea to become the "Batman", and starts his vigilante activities.
While patrolling Gotham, Batman discovers a heist going on at the chemical plant by Jack Napier and his gang and intervenes. Jack speaks to Batman and once again recites the phrase :
"Have you ever danced with the devil in the pale moonlight? I ask that of all my prey. I just like the sound of it."
This identifies Jack as his parents murderer, causing Batman to fly into a rage attacking Jack and his associates. During the fight he knocks jack criminal Jack into a barrel of chemicals. This bleaches Jacks face makes his hair turn green and gives him a disfiguring grin.
This unhinges Jack turning him into the psychotic alter ego "the Joker". To complete the transformation, the Joker dons clown make up, eventually using the clown theme to run through all of his activities and associations.
If you analyse this story it's quite clever. Effectively Batman creates his own nemesis the Joker through attempting revenge on his parents killer. This teaches the Batman that he should only be an implement of the law and not the executor of it which is why he associates himself with the Police in Gothom City, handing criminals to them rather than becoming judge, jury and executioner.
I believe this plot was similar in nature to the comic book version.
As an interesting development to this story, 2000 AD comics character Judge Dredd, is similar in nature to Batman, but he is Judge Jury and executioner. A slightly modified view of the same angst character type.
There are many stories about his orgin, but having mental disorders, it's unlikely even he would know. Jack nicklson portrayl depicted the joker as a criminal who wennt insane after being dumped into a vast of chemicals, and having his wife die the same day. In heath ledgers take, the joker has stories to accomadate his situations, involving an abusive father and self mutalation over an incident involving his wife. His name and orgin remains unknown

The difference is the joker is crazy and bad and the robin is good and robin helps batman defeat the joker.
