If your roof is leaking due to a loss suffered from a covered peril under your policy such as fire, wind hail etc. (see your policy for covered perils) then your roof leak as well as any resulting damage should be covered under you home insurance policy.

If you roof is leaking simply because it is old and has reached or or surpassed it's life expectancy (an expected and normal home owners maintenance issue) the roof leak would not be covered.
