In today's internet pop culture, it's a reference to the Japanese character called a "chouon," which is a vowel (or consonant, as well, in America) extender.

(Ex: Whyyyyy = Why~ or Kawaiiiiiiiiii = Kawaii~)

I make use of this character at the end of sentences, however, to denote a cheerful or sing-song quality to my sentence. Extra tildes at the end of my sentences denote a drawing out of the sing-song nature of the sentence or extra cheerfulness. I also add further punctuation after the tilde, depending on what I'm saying. If it's a question, I add a question mark. If I'm feeling very strongly, I'll put an exclamation point. It's very flexible.

(Ex: "How are you doing, today?" "I'm just great, thanks~")
