It depends on the version.
In Red, Blue, Green, Yellow, FireRed and LeafGreen, you need to get the Silph Scope. That is obtained from Team Rocket's Hideout in Celadon City, under the Game Corner. When you have the scope, go to Lavender Town and proceed to the top of the tower. You'll find a Marowak which cannot be caught, then on the final floor, a trio of Team Rocket Grunts. Defeat them, and you will be given the PokéFlute.
The previous answer was correct for Gold, Silver, Heartgold and Soulsilver:
First you must complete the machine parts quest before heading over to the radio station in lavender town. Inside, talk to the gentleman against the wall and he will upgrade your pokegear radio to be Kanto compatible. Once this is done go to the snorlax and open the pokegear radio. Manually tune the radio to the top of the tuner and the Pokeflute channel will operate. Then talk to snorlax and it will wake up and battle you.
edit: just help the man at the tower and he will give you the pokeflute....

Unfortunately, I don't think you can. you get a pokeflute from an old man on the top floor of the tower in lavender town. when you press the A button next 2 snorlax it will come up with an option saying "play th poke'flute" select yes and snorlax will wake up and battle u, he is on LV.30. Did you by chance go through the radio tower crisis and save the director if so he gives you a card for your pokegear and it lets you tune into the kanto radio now go to the radio on your pokegear and then tune it up high until you reach a song called poke flute now play that near snorlax and talk to snorlax it will wake up and attack you watch out its level 50.

Your pokeflute has nothing to do with Mewtwo shiny or not IN ANY GAME!. here a fact though in every encounter there is a 1/8000 that the Pokemon you see is shiny.

You cant, but there is a way. In your pokegear, you should have the radio application...thing. Place the marker at the very top of the globe in the center. There should be something called "Pokeflute" playing. If you need this for Snorlax, play it while near it, then talk to it. It WILL attack you, and it is level 50, and is holding Leftovers.
