Okay, first just pick a type of deck you wish, like: Spellcaster, Warrior, Fiend, Beast, etc.
First you can choose how many cards you want in your deck; You can buy a structure deck of 40 cards, a starter of 40 cards, pick 40 cards, or up to 60 cards.
Organise your deck into spell, trap and monster cards. Say if you have 40 cards, organise into: 20 monsters, 10 spells, and 10 traps.
After, it's time to focus on your monsters: Really, you shouldn't accept cards with 100 ATK or less in your deck, unless you can combine it with the 'Buster Rancher' equip spell card. You should keep it only if it has a good effect.
Take 4 star monsters with good A
TK and DEF, you you can build a strong defense, and offense at the same time.
Then move on to Tribute monsters. Tribute monsters are cards of which you need to send cards to the graveyard in place of it.

Place some 5/6 star monsters and only a few POWERFUL 7/8 monsters in your deck.
Now it's time for Fusion monsters. Fusion monsters are used when you need to sacrifice CERTAIN cards for an overall powerful monster. If you have fusions, keep 1 OR 2 'Polymeriation' spell cards in your deck. Otherwise you cannot summon the fusions unless you have XY Dragon Cannon, or similar card.
Spell cards: These are the cards which you should place in your deck to help you such as: Increasing your life points, summoning a powerful monster, and taking your opponents life points. Place cards in your deck which give you that advantage.
Trap cards: These cards should help you when you're in a tight spot:
they can negate attacks, quickly play cards from your graveyard etc.

And much more. Most of these cards are hard to obtain but are NOT compulsory. All I'm saying is that they're quite handy.
And that's all really, with this advice you should have an advantage over your opponent and win plenty of duels. The odd few losses are okay, but if you challenge DIFFERENT people and lose and lose and lose, then try rebuilding your deck, and follow this advice again.
