Just play though the game and once you need to go into Division 47, it will be open! It would be a waste of time just going back and forth!!!

You go to the Kine Society, and then there will be a clip showing a giant cow rising out of the floor. The Kine Society leader will tell you to go to Division 47, and then it will be unlocked.If this isn't happening, then press select and use the left and right buttons until you get to the "Missions" page. Finish any mission that you see there.
