You need to migrate Mewtwo from an earlier game in the Pal Park.
You can capture Mewtwo in Pokemon FireRed and LeafGreen then with FR or LG in the GBA slot you start diamond and a new option should be in the menu before you start the game for you to migrate from FR/ LG (max of 6 Pokemon per GBA game per day, the Pokemon can hold items however can not know any HM moves).

Put the Gameboy game (Ruby, Sapphire, FireRed, LeafGreen or Emerald) in your DS and migrate. Transfer Mewtwo from the games via Pal Park (Route 221). Mewtwo will be wandering around in grass areas inside the Pal Park.
If you want Mewtwo in Pokémon Diamond, you have to trade or migrate it. Mewtwo can be found in the Cerulean Cave in FireRed/LeafGreen and HeartGold/SoulSilver and in the Unknown Dungeon in X/Y. You can either trade it with someone else, or from the DS games and you can migrate it from the GBA games in Pal Park. You can only migrate once every 24 hours and when you did, you can find it in the Field area.
