Yes they are in love, Yusei proved that on countless occasions. He entrusted her with Stardust dragon, something he hasn't entrusted to anybody else in his entire life. He also was enraged at Divine for using her and came to her rescue in her fight against Misty by revealing Divine killed her sister and not Aki. Also, Martha pointed out that he likes her and he had nothing to say to that but attempt to shut her up out of his shyness about it. Then comes him personally teaching her how to ride duel and had her around the waist at one point in the episode. Then he caught her before she fell and she blushed and he smiled. Now for proof she loves him. When Yusei was captured she was the one who went to go and save him, she risked her own life for him as he did for her on countless occasions. She also thinks about him a lot, and whenever Yusei is in danger she knows it automatically, when her mark shines, she knows when Yusei is dueling while she doesn't know who the other signer is that is dueling. Proving she and him share a special connection, which means they're in love, their feelings became one. And when her birthmark shines Yusei's mark shines directly after it, and at one point, it was only his shinning when her's was shinning in her duel. Proving further those two have a special connection. In the upcoming episodes those two are speculated to admit they're love, in the Arc, aki duels Sherry for Yusei, something is definitely going to come out of that.

Akiza does love Yusei because in episode 71 she gets all emotional and mad when Yusei is kidnapped and Jack says "This Yusei business has you all wound up. If I didn't know any better I'd say you were in love with him." In episode 60 Sayer, disguised as a police officer, tells Yusei "I take it you have special feelings for this girl. Why else would you come to a place like this?" (this is only in the english version). The biggest thing is in the last episode of the japanese version, where Akiza ALMOST tells Yusei she loves him, but at the last second changes her mind. However, Yusei likely knew that Akiza loved him, by his reaction and returning statements in the conversation, most likely also held feelings for Akiza.

It is know that Akiza loves Yusei, as defined by Jack in episode 71 English version, but if Yusei loves Akiza then it is currently just guessed at this point. It's very clear after her duel with Yusei, when Divine had her under control she heard Yusei's voice and was snapping back to reality, and her and Yusei went on a date in episode 75 (sub version, which is the official version) where he holds her by the waist, catches her before she falls in which she blushes and he smiles. It's clear something is going to happen between them.
