Well if you mod the game I think so. Not normally though. If you have the game on PC you can mod it to allow you to play as any character you wish. e.g. You can change Excella Gionne to be Sheva. So in Story Mode Sheva will play the part of Excella. Or Warrior Chris could play the part of Wesker etc. Also, you yourself can play as any of these characters in Story.

If you mean the story line of the game, no you can't use Wesker. But you can play as him in Mercenaries, to unlock him you need at least 4 star ranking in the Water Island stage.

Nope you can't (sadly). If you do see people supposedly having "Jill" for story mode, you can be sure it's a mod. It only works for PC I think and it still displays the name "Sheva" at the bottom right screen. Yes you can play as Jill in the RE5 story, you can also play as Wesker but this can only be done by modding or finding a modded save. It works both PC & console's. You can only play Wesker in the mercenaries. It's pretty sad. It would be cool to play the game from his perspective though.
