Yes, it is possible. However, that is a very basic program and I would recommend something more advanced such as paint.NET or GIMP. Paint.NET is more user-friendly to those who use windows paint, but GIMP is more advanced. Just remember, the default texture size for each block in the texture is 16x16 pixels.
Download the Texture pack it should be in a .zip file (you will need 7zip or WinRAR) then you need to put the .zip file in your Minecraft Texture Pack folder which is located in you AppData if you are using Windows Machines.
First open the Minecraft texture template with a program that supports transparency. Either select the lava texture(found in the lower left corner of the template), and delete it; or paint over the texture with transparent color (The method you use depends on what program you are using). Finally, save the finished template as a file format that supports transparency, such as GIF or PNG.
You can use pretty much any software, however GIMP and Photoshop are considered two of the best. Microsoft Paint does not support transparency, so expect for everything to be surrounded by white if using Paint.

In the run bar type in "%appdata%" no capitals. Now open up the .minecraft folder, grab the texture pack file and drag it into the texture pack folder that's in the .minecraft folder. Now open up minecraft and press texture packs and click on your new texture! Done!

Most likely, you have installed a texture pack that is Hi-Definition or was made using Microsoft Paint. Either way, you will need MCPatcher in order to correct the textures so you can see them correctly in game.
