There is no reason for the doctor to not be played by a woman. Not only can the doctor be a woman, but Doctor Who, in an episode, has been shown to have a daughter by cloning. This implies that it is possible to change The Doctor's gender, as cloning usually yields the same gender as the original person.
After the latest regeneration transitions, the doctor takes stock of his body parts and configuration indicating that there was a possibility that the doctor didn't have to be in human or humanoid form.
On the other hand, portraying the doctor, a dominant personality, as a woman may be regarded as offensive to some cultures, though acceptable by others.

First off, please note that the Doctor is a Time Lord. Some get confused and think that all time lords are called "The Doctor" or that there is several "Doctor" Time Lords. But, in fact, there is only one Time Lord known as "The Doctor".
The Gallifreyan children who want to become a Time Lord, must first get into the Time Lord Academy and pass it. When they do pass it, they get a Tardis and a 12 count regeneration cycle. Then they also get to pick a Time Lord name. The Time Lord we watch on the TV show picked the name "Doctor".
Now since that has been established, the answer you seek :
The Doctor was born a male and has gone though one entire 12 count regeneration cycle as a male. In fact, he is the ONLY Time Lord in the entire history of Time Lords to get TWO 12 count regeneration cycles. Peter Capaldi is the actor who is playing the new 1st doctor of the new regeneration cycle. No Time Lord, good or bad, has ever changed sexes during a regeneration.
To date "The Master" is the only one who has done so and HOW he did that (changing into a Missy) has yet to be answered. However it is assumed by fans that he did it during a regeneration. But before the Master/Missy's reappearance it was assumed that the Master was killed off. The Master had been killed twice already and brought back to life with unnatural means. The first time by the Time Lord Government because of the Time War with the Daleks, and then by his human wife and cult followers on earth. Was he brought back to life again? And in doing so, did it mess him up and he became a female by mistake? These kinds of questions have yet to be answered.
Can the Doctor become a female naturally? So far on just the facts within the Doctor Who Universe, the answer is a no by facts and history. However, the show runner of the TV show in reality, Moffat, said that the Doctor can and should become a woman for at least one regeneration. But the truth is Moffat lies and he lies a lot. Will Moffat and team show how the Doctor can become a woman? It is as simple as a choice in regenerating or is it unnatural like with the Master? The Doctor has died once during the Time War as the 8th Doctor, but the cult witches of the "Sisterhood" brought him back to life and gave him a drink that turned him into the Warrior (War Doctor) and not himself as The Doctor. Maybe a drink has the ability to change a male Time Lord into a woman Time Lord. Only "time" will tell! (Pun intended)
But my question is this, those who change their sex, do so stating that they were really the opposite sex stuck within the wrong body. Now since the Doctor (a Male), has gone though 13 lives as a man, if it was just so simple to do so with a regeneration then why hasn't he already changed? To me, he likes being a male and doesn't feel trapped into the wrong body. So why would he want to change into a woman(?)
