It depends on what year(s) you are talking about. In all of Harry Potter's school years, Professor McGonagall was the Headmistress. Except for Harry 5th year, when Dolores Umbridge became Headmistress but was fired shortly after.

In the Harry Potter books, Albus Dumbledore is the headmaster and Minerva McGongall was the deputy headmistress. Twice during the series, McGongall became acting headmistress, once in the 1992-1993 school year when Dumbledore was removed by the Board of Governors and once in the 1996-19997 school year after his death.
