The Volturi had their eyes on Alec and Jane (biological siblings). It was suspected that they were doing witchcraft, so they were going to be burned at the stake. That's when Aro stepped in. He turned them both into vampires and welcomed them into the Volturi clan. Also I'm assuming that they were changed into vampires sometimes far in the past (ranging from 1400's to 1700's seeing as they were going to be burned at the stake).

Werewolves are the only thing strong enough to destroy a vampire, besides another vampire. Vampires are basically the only thing that can kill a werewolf because the venom is extremely poisonous to them and kills the wolf instead of transforming them into another vampire. Caius realizes how powerful the Quileute pack is, and that is why he is "afraid" of the wolves.
