Harley Quinn (play on the word harlequin) first appeared in "Batman: The Animated Series" epidsoe "Jokers Favor". Since then she has appeared in many other series including comic books. Her first comic book was "The Batman Adventures: Mad Love", but it was written and drawn by Paul Dini and Bruce Timm (who were 2 producers in of the animated series and Harley's creators) the comic book revealed Harley's origins as an Arkham Asylum psychiatrist who fell in love with the Joker. The story was widely praised and won the Eisner and Harvey Awards for Best Single Issue Comic of the Year. Since then she has appeared in animated series such as "The Batman" and a small cameo appearance at the end of "Batman Beyond: Return of the Joker"

The Joker was the nemesis of Batman. Nemesis means that The Joker was Batman's enemy in the many movies of Batman.
Many times (In the movies and TV shows) Batman has threatened to kill The Joker. He has never carried out his threats because it would violate his conscience. There are many comics where Joker does die and I believe there is an alternative comic called 'The nail' where Batman kills The Joker.
Ceasar Romero was the first joker in a TV series back in the 1960's, but the Joker's character was half based on Conrad Veidt's character in The Man who Laughs back in the 20s, & many concider him to be the first.

Ask a 5 year old and the answer is Shrek. Ask a 5 yr old's parent, they will also say that. It is the environment of your family, lifestyle which dictates this answer. But the Joker from the comic books and the Batman series of the 60's will come to mind for many. Not necessarily the now day serial Batman movies.
The Joker would be known by many comic fans who are also SF fans, but many Star Trek fans would only have peripheral knowledge of the Joker or other Batman characters. As both Spock and the Joker have appeared in several high-profile movies, they would both be known to many non-fans. Spock, however, is something of a cultural icon in the US, and despite having been around for 3 decades less than the Joker, would probably be considered more historically famous.
There have been many rumours from Man-Bat to Riddler to Penguin to even the Joker. However, it is too early to tell which one it will be.
