The "Clucking Cock" mission is one of two side missions CJ can do, they take the form of triathlon races where he swims, cycles then runs in a race against 11 others. The name of the missions (the other one is found up near Fisher Lagoon and is called "Beat The Cock" I am sure) comes from the fact that one of the people you race against is dressed in a chicken suit. I think the reason you are getting the "not ready yet" message is because you have to have increased CJ's stamina, swimming and cycling abilities each to a certain level before he is capable of entering the races. Alternatively, each of these races is only available on one particular day of the week (I forget which days in each location, sorry), so it may be that you are visiting the mission startpoint on the wrong day.
