a pilgrim is one who travels for religious purposes while a colonist is one who travels to start a colony. In 1607 Virginia was founded as a colony and a group (a lot) of Separatists (dedicated Puritans) negotiated with the Virginia Company of London (who held the charter for the colony) to let them live in Virginia to escape religious troubles in England. These Separatists loaded onto the Mayflower in hopes to end up with the other colonies. They sailed for 65 days at sea but missed their destination and ended up a Plymouth rock in 1620.

A colonist refers to any person who settles in a new colony or territory. Pilgrims specifically refer to the group of English settlers who founded Plymouth Colony in Massachusetts in 1620. All Pilgrims were colonists, but not all colonists were Pilgrims.
