All thing being equal, a reliable shotgun (12 or 20 gauge) is the best home defense weapon. Why? It's relatively easy to shoot. There are MANY ammo choices to suit almost any situation. It is extremely intimidating--so much so that you'll likely never have to use it.
But here come the caveats:
If you are unable or unwilling to purchase gun safe large enough to store a shotgun safely, don't get one. An unsecured firearm is more likely to be used accidentally than in a home defense situation.
If space is an issue, get a handgun and a lockbox. Don't get anything smaller than a 9mm. If you don't think you'll practice very often, get a double action revolver.
Last, get a flashlight. If you opt for a shotgun, it must be mounted to the weapon. If you use a pistol, make sure you practice shooting while holding a flashlight.
Some weapon specific suggestions:
Beginner: Remington Police 18" 12-gauge shotgun (Inexpensive, reliable, intimidating)
Advanced: Benelli M-4 12-gauge shotgun (SuperAwesome-but it's expensive and requires more practice to operate.)
Beginner: All Glocks with a calibre greater than 9mm. (Inexpensive, reliable, easy to use)
Advanced: Sig Sauer 229 in 357 SIG or any 1911 from a reputable gun maker, eg, Kimber, Colt, et al. (357 is expensive, but has a hell of a wow factor...the 45 isn't called 'Ol Ugly for nothing. Either will serve you well.)

There is no best gun because it depends on too many factors. The best gun for you is whichever gun that fits your hand the best so you can shoot it accurately. Beyond that, a gun for everyday carry must be reliable and be of an adequate caliber for self defense.
I had one on my home defense gun and hated it. I recommend staying with the factory stock.
