The postal service uses machines to sort its mail. If you are sending your mail via first-class you probably don't need to be concerned with it. However if you are doing large bulk mailings you need to be concerned, not only with the "machinability" or quality of your envelopes, (weight, size, etc) but also the placement of your address on the envelope, and the placement of the postage, and etc.
The USPS has all of the standards on their website, but to be honest, I have never found their website very helpful because it is SO CONFUSING and there is so much of it. I would go to your nearest business mail facility and ask them. While you are there you can also get the mail standards unit phone number and you can call them with your questions.
