A regular guitar amplifier is generally just the amp head itself (no speaker box), while a combo amp contains the amplifier and the speaker all in one containment box.
Sure, but you better check the output ratings on the stock radio against what you need to run the box, if it is self powered then never mind. Go to ( the12volt.com) and copy the radio wiring diagram for your radio, splice into the rear speaker wires and connect your box. Done, enjoy. Try to maintain polarity on the new box, negative to neg, pos to pos, and keep it that way, it could cause damage when you increase amperage if you don't.

I think you mean a port? No it does not need a port
I have made a talk box before. What you need is some surgical tubing, a high current dual pole single throw switch, a small - high powered- speaker and a small sealable box to fit it in. You will also need an amplifier that has the speaker output that you can use. You need to mount the speaker and the switch into the box - wiring the speaker so when you push the switch, it will route the signal to the speaker in the box or back to the speaker from the amplifier. Make sure that the box has a hole in it that you can attach the surgical tubing that is on the same side as the front of the speaker. Attach the tubing to the box. If you want, there are plumbing fitings that you can buy from your local hardware store that will allow you to remove the tubing from the box. Then seal it all up. You can use silicone for that. run the surgical tubing from the box to the microphone that you are using. The sound will travel through the surgical tubing and into your mouth and then through the microphone.
Easiest wasyis to build a regular speaker box built to the speakers specifications, and cut a hole in the box where ever you want the plexigalss. to install it you need to cut a small grove along the hole you cut earliers so that the plexiglass can sit even with the rest of the box. after that carefully place clear silicone on the surface you will place the plexiglass on and stick the piece on. pre drill holes so you can place small srews in it for added structure.
Yes it won't play unless you have a DVD player connectors on the speakers.

The box turtle can hold it's breath for nine days. Seriously.
In order to punch a hole in a box, simply place the cardboard box in front of you and punch into it with all your force. This will usually create a hole.
Depends on the speaker manufacturer and the type of box, but... Sealed box- .75 cubic ft. ported box- 1.5 cubic ft.
Typically, the speaker cabinet is powered by the Amplifier. It's the amp that is raising the volume and power to make the speaker move. "Combo" amps have the amplifier and speakers in one box.
How about this: What's inside the box? How Sound Wave Intensity changes in a speaker box to outside.
