Mister Burke can be found at Moriarty's Saloon in Megaton, until you either prime Megaton's bomb for him, or disarm it. He will then leave and head to Tenpenny Tower, where he can be found on the same floor as his room there. It is possible for him to die en route.

The only ammo you get is where you get the Alien Blaster. But apparently there is more ammo near a truck where a Mr. Handy is. Hope that helps. :)

Yes in fact it does! The same exact thing goes for Mr. Tenpenny. Wait until after you save Megaton though. Kill them both on the outside balcony of Tenpenny Tower. This keeps the Megaton settlers from attacking you, and if you are on the balcony it will keep anyone that's at Tennpenny Tower from attacking you.
