Depends on what you mean.

In a very real sense The setting of Charlotte's web is E. B. White's barn in Maine (this is where he got inspired to write the book).

If based on the book it takes place in Mr. Arable's farm and Mr. Zuckerman's farm and happens during spring, summer, autumn and winter.

The majority of the book was the Zukerman's farm where Wilbur lived. But the farm where Fern lived and Wilbur was born was the setting for the first few chapters. Then they went to the fair where Wilbur met Uncle and at last back to Zukermen's farm where he lived the rest of his days without having to worry about being somebody's meal.

The setting of Charlotte's Web is mainly on a farm, in a barn. There are a few scenes at the fair grounds though.
