No. Immigration check points aren't between cities; they're at the borders between the US and any other country. If you're concerned about road travel from Houston to Austin without a passport or other reason, you can relax, at least as far as going through immigration is concerned. If you're traveling under a foreign passport and are flying between Houston and Austin, you may be asked to show your passport upon checking in at the airport but it's for identification, not immigration.
