There were lots of informal organizations, especially around the Midwest, but the NFL was the first serious attempt at organizing a professional football league in the United States.

There have been several rival leagues calling themselves the American Football League throughout the years, but none of them predate the NFL. The best-known and most successful AFL was the one that formed in 1960 and merged with the NFL in 1970. The AFL brought the Dolphins, Jets, Bills, Patriots, Bengals, Titans (then the Houston Oilers), Chargers, Broncos, Chiefs, and Raiders into the league.
