I just got back from driving to south America from Alaska.
The only issue is the Darien Gap between Panama and Colombia - there is a section of jungle a few hundred kilometers long with no road.
So, what you do, you ship your vehicle from Panama to Colombia and take a boat yourself or a plane for an hour and then pick up your vehicle a couple days later when it arrives. Then, off you go...
