Ambien is a sleep medication called Zolpidem and is intended for the treatment of insomnia. It is a nonbenzodiazepine sedative hypnotic and does not provide false positive screen results of the standard enzyme multiplied immunoassay technique (most often used for drug screening in urine) and does not confirm positive for Benzos in confirmation by gas or liquid chromatography.

It shouldn't. It may, however show up as a sedative/hypnotic. If you've a valid prescription for Lunesta, say so up front. If you do not have an Rx for lunesta, it is a federal crime (a misdemeanor) to consume it.

Lunesta is not related to benzodiazepines and will not be reported as such on a drug screen. However, the drug screen panel may look for chemical compounds such as Lunesta. As long as you have a valid prescription, it shouldn't be an issue.
