Depends on the drug, but approximately 35-60 minutes to give it a good shot. The glycerin melts very quickly, and then the mucosa of the anus has to absorb the drug. Fr33kman, MD I have found that by making sure the glycerin suppository is pushed high into the rectum, I get relief in 5 to 10 minutes; the longer I hold, it seems like, the better it works. I like the glycerin ones because once you go, theres NO more going. Ducolux suppositories are different; they make your bowels move in 15min to an hour..but usually I get after results usually at a very bad time..

My Dr told me to urniate before inserting, wash my hands and lay still on my back for about 30 mins to an hour after inserting. Then you can resume normal activity including going to the bathroom
