Depends on the drug, but approximately 35-60 minutes to give it a good shot. The glycerin melts very quickly, and then the mucosa of the anus has to absorb the drug. Fr33kman, MD I have found that by making sure the glycerin suppository is pushed high into the rectum, I get relief in 5 to 10 minutes; the longer I hold, it seems like, the better it works. I like the glycerin ones because once you go, theres NO more going. Ducolux suppositories are different; they make your bowels move in 15min to an hour..but usually I get after results usually at a very bad time..

The length of time it takes a suppository to dissolve depends on the type of medicine. In most cases a suppository is made to dissolve within a few minutes at most.

A suppository is just a method of drug delivery. The length of the drug's effect depends on the drug. Ask your pharmacist for advice specific to the medication you're using.
