Well, in the episode where eggman awakens shadow at the end, there is a scene where knuckles starts beating up rouge, and when he flings her against a tree, she looks as though she's about to cry.

Not that I've seen, Shadow usually keeps more to himself, often blanking the world out in the process. So, I'd think not. Maybe off the scenes, yes, but not on film. Besides, Rouge usually flirts with Knuckles, Sonic, and Tails, to make THEM blush. Ecspiecally Knuckles. Don't forget that she kissed Tails in Sonic X.

If I were to guess, I would say he has a small crush on Rouge but would not be devastated if she were to reject him. Knuckles has a secret crush on rouge but rouge has a secret crush on him too.
