In Book 1: Bella, there are no antagonists to speak of. Although, as Jacob is against the marriage and the sexual relations Edward and Bella are to have on their honeymoon, he could be viewed as an antagonist, although he is not the direct opposite of the main characters in the novel.
In Book 2: Jacob, the wolves can be seen as the antagonists as they are seeking to kill the Cullen's and Bella's unborn hybrid child.
In Book 3: Bella, the antagonists are the Volturi, who have sought to destroy the Olympic Coven (Cullen family) for quite a while, and use the birth of Bella's child as an excuse to destroy them.

Vampires are basically the only thing that can kill a werewolf because the venom is extremely poisonous to them and kills the wolf instead of transforming them into another vampire. Caius realizes how powerful the Quileute pack is, and that is why he is "afraid" of the wolves.
