No, and not even close. Buddy Holly and the Crickets performed on the roof of Womble Oldsmobile (a local car dealership) in their hometown of Lubbock TX, years before The Beatles. And even they likely weren't the first to play on one.
The Beatles planned to complete their Get Backdocumentary (later re-edited and released as the movie Let It Be) with their first live concert since their Candlestick Park show in 1966. Plans for the concert went awry, and it was ultimately decided to just play a show on the roof of their own office building at Apple, for whoever could hear them, to finish off the documentary. (They played another set the next day in the Apple basement, but this footage was put first in the final edit.)

They wanted to play somewhere, but didn't want to play live in the conventional sense as they were through with all the screaming, and couldn't be where there could be open public access. Where is near by, where anyone passing by can have 'public access' but no-one can get to them and hassle them? A roof!!! It was another random, but iconic Beatles thing. Sadly one of the last.
