His chest is covered in jewels from lying on his stolen treasure for such a long time. Bilbo notices that there is a single spot on his chest not covered in jewels. He sends a message to the men of lake town telling them of this weakness. He is then shot with an arrow in this spot which ultimately leads to his demise.

Smaug's weakness is that on his left breast he has a small gap that isn't armed and Bilbo found it when he snuck into the secret entrance to the cave and he saw Smaug sleeping.

Hope this helps!

Bilbo discovered a weak point in Smaug's scaly armour, which ultimately caused his death.

In the movie adaptation, the weak point had been created by Bard the Bowman's ancestor, who tried to kill the dragon with a giant bow called a Windlance. The arrow had hit its mark, dislodging a nigh-inpenetrable scale, which later allowed his descendant to fire a killing shot into the dragon's heart.

You mean physically? He has a bare spot in his armor, on the left side of his chest (nearest his heart) and he does not realize it.
