Matt didn't write, direct, storyboard or even come up with the title of the first Simpson episode, so if you want to get technical, Matt didn't create the first episode, not in the same way Seth MacFarlane made the first Family Guy or Trey Parker & Matt Stone made the first South Park.
However it most likely would have taken the same amount of time it does now, 6 to 8 months to create the first episode.
Quite possibly a little less due to the rather crude nature of the animation and drawing style of the 1st episode. Once they got accustomed to the character & show designs it probably went faster.
