its just a relatively non toxic detergent that helps wash the blot and remove any proteins that are non-specifically bound. some people follow up the TBST washes with a straight TBS wash to remove it, but it is not necessary. some people use TBST to dilute their Ab, too. Then again, some people use TBS, and add a little BSA (like .5 or .3%) to kind of keep blocking as they go along with the steps of the process. for that matter, you can just use a weak PBS with a 1% tween to dilute your Ab, too. It probably will not matter.

A blotting paper is used to absorb excess oil from your skin.
I heard that The Queen uses black blotting paper in order that no one could read what she had written. I wrote to her to ascertain the veracity of this practice and received the following return from Miss Michele Edwards who is an Information Officer in Her Majesty's Press Office. Thank you for your letter of 22nd March. "The rumour about The Queen using black blotting paper seems to surface fairly regularly. There is no truth in this; Her Majesty uses ordinary bloting paper." ... more, not relevant. Yours sincerely etc.etc. I hope this answers you query.
it is a labours process it is timeconsuming process it required more amount of dna require 3microliter dna per sampel it is costly
A western blot is done by immobilizing cellular proteins on a thin membrane (usually PVDF or nitrocellulose) and detecting them with antibodies to specific proteins. Briefly, cells are lysed, proteins are resolved on a gel, then proteins are transferred to a membrane, and finally the proteins are detected by enzyme-conjugated antibodies.
