Your DIY 12V power source is shorting out. Something wasn't assembled correctly. Take it apart and do it over. Defective phone charger?
If you are talking about a car's cigarette lighter then yes if you have the proper phone adapter/charger.
Check the outlet by plugging in a 12v accessory (radar, gps, phone charger). If it has power, then the problem is the lighter itself. The are only a few dollars.

Check the fuse. If you use a phone charger or any other aux device in the cigarette lighter most likely blew it.

No, portable cell phone chargers will not harm your phone. I've used wall chargers and cigarette lighter chargers with phones and never had any problems related to them.
The cigarette lighter / pwer socket is a constant hot socket. In most cars you can use this power socket at all times. It is best to remove anything(except the cigarette lighter) from this socket when the car is off.
To charge a cell phone you will need either an outlet plug-in or car charger compatible with your particular model. The easiest way I've found to fix a lighter, is to replace the element ... the part that gets hot. They fail fairly often. You can find them at parts stores. Before that, however, you should check the fuse. If the fuse is good, try plugging a cell phone charger into the socket to see if the socket is being powered or not. If the cell phone charger works, go back to square one and replace the lighter element. If the charger doesn't work, you'll have to replace the lighter socket.
