When the heart is in complete relaxation. The pressure in the heart is low, and blood is flowing passively into and through the atria into the ventricles from the pulmonary and systemic circulations. The semilunar valves are closed, and the AV valves are open. Then the atria contract and force the blood remaining in their chambers into the ventricles.

The highest blood pressure in the circulatory system is when the oxygenated blood has just been pumped out of the left side of the heart. This is because the left side of your heart has to pump the blood all the way from your heart, to your brain, feet, fingers, etc. It has a long distance to travel, and it needs pressure to provide a fast journey. Artery walls are thicker than those of the veins because they carry the higher pressure blood.
