According to "Clinical Pharmacology & Therapeutics" at nature.com, "...omeprazole is unlikely to increase the risk of acetaminophen hepatotoxicity..."

Therefore, it seems that omeprazole (Prilosec OTC) is safe to take with acetaminophen (Tylenol). Just make sure that you are not allergic to either medicine and make certain that you consult your doctor or local pharmacist before beginning a new antacid regimen.
