It is the most valuable gem in the dwaves kingdom
The Arkenstone is a large white gem sought by Thorin Oakenshield in The Hobbit . It was discovered beneath Erebor (the Lonely Mountain) by Thorin's ancestor, Thráin the Old, and shaped by the Dwarves .
It was big enough that Bilbo could not close his hand around it. Thorin described it : "It was like a globe with a thousand facets; it shone like silver in the firelight, like water in the sun, like snow under the stars, like rain upon the Moon!" When he first saw it he caught his breath. It appeared to have its own inner light and ". . . it took all light that fell upon it and changes it into ten thousand sparks of white radiance shot with glints of the rainbow."

The Arkenstone does nothing. In fact, the only reason the dwarves want it back is because it is a great treasure and a magnificent jewel. It was discovered beneath the roots of the Lonely Mountain and is also known as the heart of the mountain.
If you are referring to near the end of the book where Thorin rejects Bilbo, it is because of a dwarf's greediness for gold and treasure. Thorin had been searching for the Arkenstone of his father, Thrain, in the Lonely Mountain, and was upset that he could not find it. He was shocked when he found out that the very Hobbit that the dwarves had put all of their trust in had betrayed them and found the Arkenstone and did not tell him. Bottom line, dwarves are greedy. Hope this answer helped. :)
Spoiler Alert! Thorin is laid to rest under the Lonely Mountain with the Arkenstone on his breast.
The thief in Deathly Hallows can refer to Kreature, who stole the locket from the house, Mundungus, who also stole the locket, or, most probably, referring to Gellert Grindelwald since Harry witnessed the vision of him stealing the Elder Wand in the chapter titled "The Thief."
