Just a wild guess, but I would start at the top of the heads.......should see a spark plug wire connected to a plug........viola! spark plug.
#1 DO NOT try to take plugs out with engine still hot and be sure you are straight up and down on plug

The Champion RC12YC spark plug is one of the most preferred spark plugs. Victor series cylinder heads are also widely used especially by racers.
For a 2001 Toyota Sequoia, the factory and spark plug manufacturer recommended gap is .032" (.813mm). The spark plug manufacturer's recommended torque for flat-seated plugs / aluminum heads is 20-30 n-M (15-22 ft lbs), but double check your owner's manual (if available) to be sure.

Not definite, but possibly wrong timing when reinstalling cams and chain and heads, etc. other option is wrong spark plug wiring. spark plug wires on the wrong plug!
The ( 2 ) engine , cylinder heads , are on each side of your V6 engine where the spark plugs are - just look for where the spark plug wires connect to the spark plugs
I am running the exact same combination with 0.035" gap. Good results.
