It is not that people built a city along the Tiber. The foundations of the city involved the unification of several independent settlements on what were to become the Seven Hills of Rome. The earliest archaeological finds of settlements in the area of the city date to some 5-600 years prior to its foundation.
One of the favourable factors of Rome's location was the hills. People liked to live on hills because they were easier to defend from raids. As for the Tiber, Rome was by its only Ford of this river. Since the Tiber cut from the Apennine Mountains (a range which forms the backbone of the Italian peninsula) to the east to the coast to the west, this made her the main north-south communication point. The Tiber also provided a navigable route to the sea.
