In the upper room of the Temple in Jerusalem. The Savior laid his hands the heads of his Apostles and gave them the Gift of the Holy Ghost. Many gifts of the Spirit were manifest at the conclusion of this meeting.

The descent of the Holy Spirit upon the apostles is commonly referred to as Pentecost, and ocurred in Jerusalem:

While meeting with them, he [Jesus] enjoined them not to depart from Jerusalem, but to wait for "the promise of the Father about which you have heard me speak; for John baptized with water, but in a few days you will be baptized with the holy Spirit." Acts 1:4 NAB. (see also Acts 1:8, 12)

Also, during his speech at Pentecost, St. Peter widens his address to other on-listening citizens of Jerusalem, indicating that he is indeed speaking in Jerusalem:

You who are Jews, indeed all of you staying in Jerusalem. Let this be known to you, and listen to my words. Acts 2:14 NAB.

Pentecost occurred 50 days after Easter. See Acts chapter 2. The Holy Spirit rushed upon the Apostles (and maybe other disciples? I am not sure) and the Blessed Virgin. It is also the birthday of the Church.
