During World War II, Hitler's defeats were rare in the first several years of the war but quite common in the last several years. Notably, in 1940, Hitler's air force was defeated in the Battle of Britain. Several years later, in late 1942, Hitler's army was decisively defeated in the Battle of Stalingrad, which proved to be one of the major turning-points of the entire war.

You'd have to define 'first defeat'.

Hitler's land forces were delayed or outmaneuvered in a number of relatively minor ways during his advance through Western Europe, but ultimately they were victorious.

The Battle of Britain (which peaked in late1940 saw many defeats for the Luftwaffe. Those were probably the first significant defeats.

The first significant defeat of Hitler's land forces (under Rommel) was at Tobruk; a small Libyan outpost held by Australian troops and British artillery. The seige of Tobruk lasted from April 1941 to December 1941 and greatly frustrated Hitler's North African ambitions. Rommel was later defeated completely at El Alamein, in a tank battle against Australian, New Zealand, Canadian, English and Indian troops under the command of Montgomery.

Towards the end of 1941 Hitler's troops were also pushed back from Moscow which was the beginning of a much greater defeat of the Germans. It's arguable about which defeat was inflicted first.

However, the attack on Tobruk is widely accepted as the first significant defeat of German Land Forces. That occurred first in around April 1941 (and multiple other attacks failed thereafter leading to the seige)
