It depends. Because many amino acids have more than one codon, it may not affect the protein at all. However, if it does change the amino acid sequence, it could cause a change in the three-dimensional structure of the protein, resulting in a mutation.

The effects of a substitution in one of the nucleotides in a synthesis of polypeptide would cause a mutation. However without knowing which nucleotide you were substituting, and which one you were replacing it is difficult to give a more specific answer.

It would create a mutation, because if a nitrogen base is being replaced, it causes a change in the DNA which creates a mutation.
