Offset maybe be relative to several things.
In a VR engine, though, it is most likely referring to the distance between each piston's line of travel and the line that passes through the crankshaft's centre.
Offset "\ /", no offset "V"
This is also sometimes called fulcrum.
The purpose of this offset is to allow the cylinders to be arranged in a very tight angle without intersecting.
To have such a tight angle without offset (as most normal Vs) would imply extremely long conrods in order to avoid bore interference.
In the case of VW's W12 (or more correctly WR12), which is basically two mated VR6s (V-VR), the offset is 12,5mm.
