They had to leave a town called Weed because Lennie touched a girl's dress and she claimed that he raped her. George tells Lennie about their dream of owning a piece of land. -They arrive at the ranch and meet Candy, the old swamper with a missing hand. He introduces them to the boss. In the bunk house they meet Curley, the boss's son who has a virile personality, his wife who is described as a 'tart' and Slim, the jerkline skinner and unofficial leader of the men.

-George tells Slim everything about what happened in Weed. Slim gives Lennie a puppy. Carlson, a typical insensitive ranch hand, convinces Candy to let his old dog be shot. Candy buys into George and Lennie's dream with his savings. Curley comes in and starts a fight with Lennie. When ordered to retaliate by George, Lennie breaks Curley's hands. Lennie is incredibly strong. Slim tells Curley to say that he got his hand 'caught in a machine'.

-The men go to town, leaving Candy, Lennie and Crooks, the black stable buck behind. Lennie goes into Crooks's room. Crooks teases him. Candy comes in and talks about the dream. Curley's wife comes in and Crooks tells her to leave. She threatens him with a charge of rape.

-Lennie kills the puppy in the barn. Curley's wife comes in and talks to Lennie about her dream of being a movie star. She invites Lennie to feel her soft hair, as Lennie is drawn to soft things. She panics and Lennie holds on. He breaks her neck and she dies. It is obvious Lennie is the killer. Curley is very eager to kill Lennie to avenge his hand.

-George meets Lennie back at the pool where the novel started. He tells Lennie the story of the dream and then shoots him in the back of the head. He knows this is kinder to him that allowing him to be killed maliciously by Curley. Slim and the others find him there and Slim comforts him for killing Lennie. Curley and Carlson do not understand the friendship between George and Lennie.

The following morning, George and Lennie reach the bunk house at the farm. Candy, the old man who shows them the bunk house, tells them that his boss was expecting them the night before and was angry when they weren't ready for work in the morning. Near his bed George finds a can of insect poison, which leads him to think that his bunk is infected, but the old man reassures him, telling him that person who had the bed before was a meticulous blacksmith named Whitey who kept the insect killer around even though there were no insects to kill. As George prepares to meet the boss, Candy reports that he is a nice enough man although he takes his anger out on the black stable buck, Crooks. Soon enough, the boss enters and asks George and Lennie for their work slips. George attempts to speak for both Lennie and himself, but the boss notices Lennie's silence and questions him directly. Lennie attempts to speak for himself, aping phrases that George has spoken, but sounds completely ridiculous. George tells the boss that Lennie isn't bright, but that he's as strong as a bull and an incredibly hard worker. The boss wonders why George is willing to take care of Lennie; George tells the boss that Lennie is his cousin and that he promised his mother to look after him. When the boss wonders why they left their last job, George tells him that they were digging a cesspool and completed the work. When the boss leaves, George scolds Lennie for failing to keep completely silent. George admits that he lied about Lennie being his cousin. Candy returns with his old sheepdog, and George snaps at him for eavesdropping. Curley, a haughty young man, enters the bunk looking for the boss, who is his father. He behaves threateningly to Lennie. When he leaves, Candy explains that Curley, who is short, hates big guys like Lennie out of jealousy. George says that however tough Curley may be, he will be sorry if he picks a fight with Lennie, who is incredibly strong. Candy notes that Curley was recently married to a local beauty and that he has become more cocky ever since. Curley wears a left glove full of Vaseline to keep the hand soft for his wife, whom the old man thinks is a tart. George warns Lennie to avoid Curley. On cue, Curley's wife comes to the bunk house looking for her husband. She is provocatively dressed and quite flirtatious. When she leaves, George remarks that she's a tramp, while Lennie says that she's pretty. George warns him to keep away from her. Next to enter is Slim, the widely respected jerkline skinner. Slim questions George and Lennie about what work they can do. Carlson, a large, big-stomached man, also enters the bunk house and asks Slim whether his dog had her litter last night. Slim tells him that she had nine puppies, but that he drowned four immediately since she couldn't feed so many. Carlson complains about the smell of Candy's old sheepdog and tells Slim that Candy should put it out of its misery. Curley enters again and confronts George, asking if his wife has been around. George admits that she was at the bunk house. Curley seems eager to start a fight with anyone.

In Chapter 5 of Of Mice and Men, Lennie has just killed his puppy by accident and is stroking its dead body. He worries that George will be angry at him and won't let him raise rabbits on their farm. He hurls the dead puppy across the room in frustration, then retrieves it and strokes it again.

Curley's wife comes in and sits beside him. He hides the puppy and tells her he's not allowed to talk to her. She reassures him that they won't get in trouble because everyone else is busy with a horseshoe tournament.

She sees the puppy and comforts him about its death, then complains she's lonely. She tells him she wants a different life, and had a chance to go to Hollywood to become a movie star, but was forced to settle for marrying Curley.

Lennie tells her about the rabbits, and she asks him why he likes animals so much. Lennie tells her he likes soft things, and she lets him pet her hair. But he holds on too tight, which scares her so she cries out. Lennie panics, putting his hands over her mouth to quiet her, but he tightens his grip too much and shakes her until her neck is broken.

Realizing that he's killed her, he tries to hide her in the hay. He worries that George will be angry if he finds out. He flees, taking the puppy's body with him. He runs to the clearing in the woods (where the novel began).

Candy discovers the body in the barn when he goes looking for Lennie. He calls George, who realizes what's happened and hopes that Lennie can just be locked up where he can't hurt anyone but can be treated well. However, Candy tells him that Curley will have him lynched.

Candy asks George if their dream of buying the farm will ever happen, but they both know it won't. George always knew it wouldn't, but Lennie had liked the idea so much that he went along with it.

Worrying that he will be implicated in Curley's wife's death, he has Candy deliver the news and pretends not to have seen the body himself.

When Candy tells the ranch workers what happened, a mob gathers. Curley demands that they find Lennie and kill him. He has them get Crooks's shotgun, and they leave to find and shoot Lennie.
