Topoisomerase: are isomerase enzymes that act on the topology of DNA

Helicase untwists the double helix and separates the template DNA strands at the replication fork. This untwisting causes tighter twisting ahead of the replication fork, and topoisomerase helps relieve this strain

It cleaves both strands of the DNA helix during DNA replication and transcription.

This relieves the torsional stress during DNA unwinding. After DNA has unwound, it helps reconnect (religate) the double stranded DNA, restoring the DNA chain integrity.

The function of topoisomerase is to unwind the helix of DNA into a regular, ladder-like shape. Topoisomerase is only found in primates. In other organisms, the molecule is called gyrase that unwinds the DNA.

This torsional stress is relieved by the enzymes DNA topoisomerases. Topoisomerases relieve torsional stresses in duplexes of DNA by introducing either double- (topoisomerases II) or single-stranded (topoisomerases I) breaks into the backbone of the DNA. These breaks allow unwinding of the duplex and removal of the replication-induced torsional strain. The nicks are then resealed by the topoisomerases.

How does topoisomerase affect the DNA strand during DNA replication?

The topoisomerase enzyme uncoils the double helical structure of DNA during its replication to form the replication fork. In eukaryotes both posive and negative supercoils get unbind by topoisomerase I & II respectively.Topoisomerase isomerase unwinds DNA to form replication fork

What enzyme is responsible for breaking the hydrogen bonds between the two strands and unwinding the helix?

Helicases and gyrases both function to uncoil the supercoiling of the two DNA strands. Moreover, gyrase is responsible for adjusting the tension in the two strands so that they don't snap while unraveling.
