I think there would still have been a CPR but it would have taken many more years to build since there were no Chinese who were willing to do hard dangerous work for less than half of what canadains were being paid.
Most of the labour used to build the Canadian Pacific Railway were immigrants because they would work for less pay. On the eastern side the largest ethnic group would have been Irish. On the western side it was the Chinese.
The Samurai were Japanese, not Chinese. The samurai ended about 1873, which would have been the Qing dynasty.
Sadly, American jobs have been outsourced to China because Chinese workers are willing to do work for extremely low pay.

Probably but not all the workers would have been slaves.The work force was most likely a mixed one of slaves and free workers. All the skilled, supervisory and administrative tasks would have been done by free workers. The slaves would have done heavy dangerous work such as quarrying, dragging the stones, putting them in place, extending and repairing the ramps used to raise the stones to the required level. A total of 20,000 to 30,000 workers would have been needed for the task.

Factory workers used to live in back-to-back houses near railway lines and the factories. They would have one communal toilet down the street that would constantly be disgusting and grubby and would have never been cleaned!
