His father king Philip II of Macedon developed a superior form of warfare and with associated bribary was able to conquer or subvert the major cities of Greece and the Amphythionic Council (which pontificated over Hellenic affairs) and get himself declared Hegemon of Greece. He then determined to defeat the Persian Empire, and began preparations and softening up action to begin this.
He was assassinated before he left for Asia. His natural successor was one of his sons Alexander (the eldest son was mentally deranged, probably a failed attempt to poison him) who had been brought up to be a war leader in his father's shadow. There are suggestions that Alexander had been complicit or at least passive in the assassination.
After Philip's death, the Macedonian army by acclamation elected Alexander as king of Macedonia, and after suppressing tentative uprisings in the Greek citiy-states, he also was declared Hegemon of Greece (hegemon meant leader, not ruler). He then proceeded on his father's plan to capture the Persian Empire with the Macedonian army, its allies and Greek city contingents.
His father Philip was assassinated and so he became king of Macedonia. But from a young age he had impressed visitors with his charm and intelligence. One of the best things about him was that he never asked his men to do anything he wouldn't do. For example, he and his army were marching through a desert region where there was little water. Some of the men managed to scrounge up a helmet full of water and offered it to Alexander. He refused it when he saw how thirsty the rest of his men were, thinking it unfair that his thirst be assuaged while the others remained thirsty.
He also inspired his men with acts of great courage. There was a battle agains the Mali and he jumped off a wall (this was a walled city) into the middle of the enemy. He survived but was wounded.
