Yes. Between December 1811 and March 1812 a series of earthquakes hit the Central Mississippi Valley (reports say they even shook residents in Washington DC). This caused the Mississippi River to temporarily change direction and flow from South to North.

No. It drains nearly due south. If it drained north, the Gulf of Mexico would drain into the Great Plains! When it's all said and done, the Mississippi River drains downhill like most rivers.
